dc.description.abstractA LC é uma doença infectocontagiosa, crônica e debilitante de ovinos e caprinos que tem distribuição mundial e alta prevalência em rebanhos causando prejuízos econômicos. O tratamento com antibióticos é pouco eficaz, devido ao agente causador, a bactéria intracelular facultativa Corynebacterium pseudotuberculosis, formar biofilmes envolvendo a lesão, o que dificulta a ação das drogas na eliminação do patógeno. Com isso, a medida profilática mais eficiente é a vacinação dos animais saudáveis, porém até o momento nenhuma das vacinas comerciais disponíveis conferem proteção adequada. Este estudo teve como objetivo avaliar o potencial imunomodulador dos três antígenos recombinantes (rPLD, rCP09720 e rCP01850) agrupados em uma única formulação vacinal associados ao adjuvante saponina para ser testado na espécie alvo (ovinos). As proteínas recombinantes, rPLD, rCP01850 e rCP09720 foram expressas de forma heteróloga em Escherichia coli e purificadas. Para os experimentos de imunização e desafio, foram utilizados 12 ovinos soronegativos para a LC, que foram divididos em dois grupos de seis animais cada e distribuídos randomicamente nos seguintes grupos: G1, inoculado com solução salina 0,9% (grupo controle); G2, imunizado com a formulação multiproteica contendo 100 µg de cada proteina. Foram administradas duas doses da formulação vacinal por via subcutânea, intervaladas por 21 dias. O desafio foi realizado 30 dias após a última dose da vacina através da inoculação por via subcutanea de 105 UFC da cepa virulenta MIC6 de C. pseudotuberculosis. Após desafio os animais foram avaliados clinicamente por 12 semanas, e ao final deste período foram eutanasiados. Os linfonodos e órgãos internos foram avaliados quanto à presença de lesões. Amostras de sangue foram coletadas um dia antes da primeira imunização (dia 0), e nos dias 21, 49, 77, 105 e 133 do experimento para detecção de anticorpos anti-rPLD, anti-rCP01850 e anti-rCP09720 por de ELISA indireto. Para a designação dos níveis das lesões foram atribuídos escores conforme a gravidade das lesões. O aumento significativo nos níveis de IgG anti-rPLD, anti-rCP01850 e anti-rCP09720 foi observado a partir do 21º dia experimental. Os valores dos níveis de IgG permaneceram até o final do experimento (dia 133). Na análise das lesões macroscópicas e microscópicas encontradas nos linfonodos maiores scores no grupo não vacinado (G1). Quanto a lesões nos órgãos foi possível observar uma redução significativa de 100 % das lesões pulmonares no grupo vacinado (G2). Assim, conclui-se que a formulação vacinal contendo as proteínas recombinantes rPLD, rCP01850 e rCP09720 pode ser uma estratégia promissora para o desenvolvimento de vacinas contra a LC, visto que, após o desafio nos ovinos contra o C. pseudotuberculosis, ocorreu redução de lesões em linfonodos e pulmões bem como a indução de resposta imune humoral.
dc.description.abstract2Caseous lymphadenitis is a chronic and debilitating infectious disease of sheep and goats that has worldwide distribution and high prevalence in herds causing economic losses to their producers. Antibiotic treatment is ineffective, because the facultative intracellular bacterium Corynebacterium pseudotuberculosis, forming biofilms involving the lesion, which hinders the action of the drug in eliminating the pathogen. Thus, the most efficient prophylactic measure is the vaccination of the animals, but the commercial vaccines available will not have adequate protection. This study evaluated the immunomodulatory potential of recombinant antigens (rPLD, rCP09720 and rCP01850) associated in a vaccine formulation with saponin as adjuvant tested in sheep. Studies with recombinant subunit vaccines were limited to testing only in murine, which can make the results inclusive for the disease in sheep, because the immune response of these animal species are of different profiles. The proteins, rpld, rCP01850 and rCP09720 were expressed in Escherichia coli and purified. For the immunization and challenge experiments, were used 12 sheep seronegative for CL, divided into two groups of six animals each. The animals were randomly distributed in groups: G1, inoculated with 0.9% saline (control group); G2, immunized with the multiprotein formulation. Two doses of the vaccine formulation were administered subcutaneously, interval for 21 days. The challenge was performed 30 days after the last dose vaccine for subcutaneous inoculation of 105 CFU of the virulent strain MIC6 of C. pseudotuberculosis. After challenge the animals were clinically evaluated for 12 weeks, and at the end of this period they were euthanized. Lymph nodes and internal organs were evaluated for the presence of lesions. The Blood samples were collected one day before the first immunization (day 0), and on days 21, 49, 77, 105 and 133 of experiment for detection of anti-rPLD, antirCP01850 and anti-rCP09720 antibodies by indirect ELISA. Scores were assigned according to the severity of the lesions. Significant increase in the anti-rPLD, anti-rCP01850 and antirCP09720 IgG levels were observed starting from the 21st experimental day, denoting the effect of the first MRV dose on the humoral immune response. The values of levels remained until the end of the experiment, representing a sustained humoral immune response induced by vaccine. In the analysis of macroscopic and microscopic lesions in lymph nodes it was higher in the non-vaccinated group (G1). Thus, using this vaccine formulation with recombinant proteins of subunit may be a promising strategy for the development of vaccines against CLA, because it significantly protected the challenged sheep against C. pseudotuberculosis, reducing lesions in lymph nodes and lungs and inducing a sustainable production of IgG.
