dc.description.abstractResumo Capítulo II Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) é uma das principais pragas da fruticultura mundial, causando danos diretos e indiretos que afetam diversas frutíferas. O uso dos fungos entomopatogênicos (FEs), é uma alternativa promissora dentro do controle biológico de pragas. Recentemente vários estudos estão sendo realizados para avaliar a eficiência dos entomopatógenos no controle de moscas-das-frutas. Neste trabalho se avaliou o potencial de quatro fungos entomopatogênicos sobre adultos de C. capitata, mediante duas formas de exposição (inoculação tópica e pela alimentação), em condições de laboratório. Os tratamentos avaliados foram Beauveria bassiana com concentração de 2 x 10⁹ UFC/mL, Cordyceps fumosorosea 3 x 10⁹ UFC/mL, Cordyceps javanica 1x10¹⁰ UFC/mL, Metarhizium anisopliae 6x10⁸ UFC/mL e água destilada esterilizada como controle. Cada tratamento foi composto por 20 repetições, sendo cada mosca (3 a 5 dias de idade) uma repetição. A aplicação tópica foi realizada em adultos (previamente anestesiados a frio), com 1 μl de cada suspensão fúngica, aplicadas no dorso da mosca. Foram avaliadas duas doses (0,5 e 1,0 L ou Kg. ha-1). No método pela alimentação as moscas passaram 12 horas sem alimentação antes de iniciar o bioensaio. A dieta de alimentação foi misturada com os conídios dos tratamentos, em duas proporções (1:1 e 1:2), permanecendo dentro das gaiolas por 48 horas. Em seguida, as moscas de ambos experimentos, foram transferidas para gaiolas plásticas (25,5 x 25 x 12 cm), com alimento e água ad libitum, e acondicionadas em câmaras BOD (temperatura de 25 ± 1°C, umidade de 65 ±10% e fotofase de 12 horas). A mortalidade das moscas foi registrada diariamente por 21 dias. Os exemplares mortos foram acomodados em placas de Petri (5 x 1cm), sobre papel filtro e um pedaço de algodão umedecido e colocados em câmaras BOD. Ao final das avaliações no método pela inoculação tópica constatou-se, que a maior mortalidade foi para M. anisopliae com mortalidades de 75% e 90% nas duas doses testadas, respectivamente. O menor valor de tempo letal médio (TL₅₀) foi para M. anisopliae (3,52 e 3,55 dias). Por outro lado, no método pela alimentação C. javanica alcançou mortalidades de 95% para ambas proporções testadas e com um TL₅₀ de 5,86 e 5,95 dias. Beauveria bassiana, apresentou um aumento de mortalidade na proporção maior (1:2), atingindo uma mortalidade de 70% e um TL₅₀ de 3,72 dias, porém as moscas mortas deste tratamento, não apresentaram sinal de micose. The use of entomopathogenic fungi (EPFs) is a promising alternative within the biological control of pests. Recently, several studies are being conducted to evaluate the efficiency of entomopathogens in the control of fruit flies. In this work, the potential of four entomopathogenic fungi on adults of C. capitata was evaluated, through two forms of exposure (topical inoculation and feeding), under laboratory conditions. The treatments evaluated were Beauveria bassiana with a concentration of 2 x 10⁹ CFU/mL, Cordyceps fumosorosea 3 x 10⁹ CFU/mL, Cordyceps javanica 1x10¹⁰ CFU/mL, Metarhizium anisopliae 6x10⁸ CFU/mL and sterilized distilled water as control. Each treatment consisted of 20 replicates, each fly (3 to 5 days old) being a replicate. The topical application was performed on adults (previously cold anesthetized), with 1 μl of each fungal suspension, applied on the back of the fly. Two doses were evaluated (0.5 and 1.0 L or Kg. ha-1). In the feeding method, the flies spent 12 hours without food before starting the bioassay. The feeding diet was mixed with the conidia of the treatments, in two proportions (1:1 and 1:2), remaining inside the cages for 48 hours. Then, the flies from both experiments were transferred to plastic cages (25.5 x 25 x 12 cm), with food and water ad libitum, and placed in BOD chambers (temperature 25 ± 1°C, humidity 65 ± 10% and 12-hour photophase). Fly mortality was recorded daily for 21 days. The dead specimens were placed in Petri dishes (5 x 1 cm), on filter paper and a piece of moistened cotton and placed in BOD chambers. At the end of the evaluations in the method by topical inoculation, it was found that the highest mortality was for M. anisopliae with mortalities of 75% and 90% in the two doses tested, respectively. The lowest value of mean lethal time (TL₅₀) was for M. anisopliae (3.52 and 3.55 days). On the other hand, in the feeding method C. javanica reached mortalities of 95% for both proportions tested and with a TL₅₀ of 5.86 and 5.95 days. One of the difficulties in controlling this species is that it completes its life cycle (pupa) in the soil. In this sense, the application of entomopathogenic fungi (EPFs) may represent an efficient alternative for control during the pupal stage. These EPFs have the ability to persist for long periods in soils, making this environment ideal for their conservation and dissemination. Therefore, the objective of this work was to evaluate the efficiency of four FEs on the pupal stage of C. capitata, under laboratory conditions (temperature 25± 2ºC, RH of 65 ± 10% and photophase of 12 hours). The treatments evaluated were Beauveria bassiana (2 x 10⁹ CFU/mL), Cordyceps fumosorosea (3 x 10⁹ CFU/mL), Cordyceps javanica (1x10¹⁰ CFU/mL), Metarhizium anisopliae (6x10⁸ CFU/mL) and sterilized distilled water as control. For this, three forms of application were used: topical method, immersion method (30 seconds) and contaminated soil, at concentrations 0.5-1.0 L or Kg. Ha ˉ¹. The experiments were composed of 20 replicates per treatment, where each puparia corresponded to one replicate. The trials were evaluated for 21 days, flies that managed to emerge were transferred to plastic cages and fed a diet based on leaven and sugar (1:4) and water. Pupae and adults were transferred to a BOD chamber until sporulation symptoms were observed. The best results for the topical method were observed for B. bassiana at the highest dose, reaching 65% total mortality in puparia, while in emerged adults B. bassiana and M. anisopliae reached mortalities of 50 and 45%, respectively. In the immersion method (30s), the best result was for C. fumosorosea at the highest dose, with a 55% total mortality. The total mortality after adult emergence reached 70% for M. anisopliae. For the method in contaminated soil the best total mortality was verified at the lowest dose, for M. anisopliae with 80%, being the 60% mortality found in the flies that presented sporulation, C. javanica the highest dose improved its total mortality, reaching 70%. The results suggest that the EPFs evaluated, present control effect on puparia of C. capitata
