| dc.description.abstract | O melão (Cucumis melo) é uma olerácea de grande importância econômica no Nordeste, pois representa 95% da produção no território nacional. Um dos elementos químicos que influenciam diretamente na produtividade do melão é o nitrogênio. Por ser um gás quimicamente inerte, poucos organismos conseguem fixá-lo na sua forma livre, onde alguns obtém através da alimentação e outros por meio da quebra da ligação tripla, seja por meio de descargas elétricas, processos industriais ou fixação biológica. Esta última, realizada por bactérias, sejam de vida livre ou simbiontes. Tais bactérias promovem a assistência na recuperação de solos degradados, redução da contaminação de água e solo e redução significativa no custo da produção. Logo, o objetivo deste trabalho foi isolar as bactérias capazes de fixar nitrogênio provenientes da rizosfera do melão. A coleta foi realizada em uma fazenda na cidade de Aracati-CE, onde 5 g da amostra de solo da rizosfera do melão foram adicionadas em 25 mL de água destilada e centrifugadas a 4.000 rpm, em seguida foi retirado 10 µL do sobrenadante que foram adicionados com auxílio da alça de Drigalski a dois diferentes meios: Nyda e Meio Nutritivo (MN) preparados em placas de Petri. Posteriormente as placas foram incubadas em durante 48h a 28-30°C. As colônias isoladas foram cultivadas em Meio Livre de Nitrogênio (MLN) para a seleção e verificação da ação de fixação de nitrogênio durante 10 dias. O resultado apresentado é de caráter qualitativo, onde das 62 linhagens obtidas e testadas, 40 indicaram positividade para fixação de nitrogênio de acordo com seu crescimento em (MLN). Realizado o teste de redução de nitrato, o teste constatou que 32,5% das linhagens fazem redução, 2,5% das linhagens reduzem de nitrato para nitrito e 65% das linhagens reduziram a outros compostos. O teste tintorial apresentou 72,5% de bactérias Gram-negativas e 27,5 % de bactérias Gram-positivas. Este resultado demonstrou que as bactérias isoladas apresentam potencialidade para fixar nitrogênio restabelecendo uma alternativa sustentável do uso de fertilizantes. | |
| dc.description.abstract2 | Melon (Cucumis melo) is a vegetable crop of great economic importance in Northeast Brazil, representing 95% of national production. One of the chemical elements that directly influences melon productivity is nitrogen. Being a chemically inert gas, few organisms can fix it in its free form; some obtain it through food and others through the breaking of the triple bond, either through electrical discharges, industrial processes, or biological fixation. The latter is carried out by bacteria, whether free-living or symbiotic. These bacteria help in the recovery of degraded soils, reduce water and soil contamination, and significantly reduce production costs. Therefore, the objective of this work was to isolate nitrogen-fixing bacteria from the rhizosphere of melon. The sample was collected from a farm in the city of Aracati-CE, where 5 g of melon rhizosphere soil sample were added to 25 mL of distilled water and centrifuged at 4,000 rpm. Then, 10 µL of the supernatant were removed and added, using a Drigalski loop, to two different media: Nyda and Nutrient Medium (NM) prepared in Petri dishes. The plates were subsequently incubated for 48 hours at 28-30°C. The isolated colonies were cultured in Nitrogen-Free Medium (NFM) for selection and verification of nitrogen fixation activity over 10 days. The results presented are qualitative, showing that of the 62 strains obtained and tested, 40 indicated positivity for nitrogen fixation according to their growth in NFM. The nitrate reduction test showed that 32.5% of the strains reduced nitrate, 2.5% reduced nitrate to nitrite, and 65% reduced it to other compounds. The staining test showed 72.5% Gram-negative bacteria and 27.5% Gram-positive bacteria. This result demonstrated that the isolated bacteria have the potential to fix nitrogen, re-establishing a sustainable alternative to the use of fertilizers. | |
